磷饥饿诱导水稻根表铁膜形成机理初探

The mechanisms of iron plaque formation on the surface of rice roots induced by phosphorus starvation

  • 摘要: 采用溶液培养的方法,初步探索了磷饥饿诱导水稻根表铁膜形成的机理。磷饥饿24h后水稻的根表出现了明显的红棕色物质的沉积,扫描电镜的能谱分析结果显示,红棕色物质是铁的氧化物。针对这一现象,首先研究了没有水稻生长的正常磷营养液和缺磷营养液的变化,结果表明二者之间全波长的扫描图谱没有出现差异。采用酸碱混合指示剂的琼脂染色方法,观察了水稻根系表面及根际pH值的变化情况,并分别测定了正常磷营养(P)和缺磷(P0)2种条件下水稻的根系活力。结果看出,缺磷时水稻根系活力高于磷营养正常的处理,尤其是基因型Jin23A,其P和P0处理间根系活力差异极显著。水稻根表三价铁的浓度高于二价铁,并且缺磷根系表面三价铁和二价铁浓度均明显高于供磷处理;缺磷处理水稻根质外体沉积的铁浓度也明显高于供磷处理。因此,初步确定磷饥饿诱导水稻根表铁膜形成是生物学基础上的化学反应过程。

     

    Abstract: The possible mechanisms of iron plaque formation induced by phosphorus (P) starvation for rice seedlings were studied in solution culture or agar culture. Rice roots appeared reddish after 24 h in nutrient solution without P, which might indicate that iron plaque was induced on the surface of rice roots; however, they remained white in solution with P. The rice roots growing in medium with P and without P were explored with Scanning Electron Microscope in accord with EDAX (SEM/EDAX) to confirm Fe existence in reddish materials on the root surface of rice. The results showed that there was iron in the reddish materials accumulated on the surface of rice root with P starvation. Iron plaque formation induced by P starvation was an interesting phenomenon observed during the experiments. A series of experiments had been done to find the possible mechanisms of iron plaque induced on the roots without P application. The value of pH and Eh in rhizosphere might be two main factors to induce iron plaque formation on the rice root surface. Furthermore, the changes of pH on the surface of rice root in agar with P and without P were investigated using pH indicators. The pH was lower in P0 treatment than that in P treatment. However, the oxidation of α-NA of intact rice root of P starvation for three genotypes was significantly stronger than that of P treatment. It suggested that P starvation enhanced the oxidizing ability of rice root. The results also showed that Fe concentrations in apoplast of root without P application were significantly higher than those with P treatment, and Fe (Ⅲ) concentrations were much higher than Fe(Ⅱ) concentrations on the surface of rice root. Therefore, the formation of iron plaque on rice root induced by P starvation might be the results of bio-chemical reactions occurred on the rice root surface between rice root and iron in nutrient solution.
