高压液相色谱法同时测定植物组织中六种细胞分裂素组分和生长素

Simultaneous determination of six endogenous cytokinins components and auxin in plant tissue by high performance liquid chromatography

  • 摘要: 利用高效液相色谱研究了植物组织中六种细胞分裂素组分和生长素含量的测定方法。结果表明,采用反相色谱柱Hibar.RT250-4.6,在45℃恒温下以甲醇1%乙酸(40/60,v/v)溶液为流动相0、.6.mL/min等度洗脱,在=269.nm处能准确检测出植物组织中六种内源细胞分裂素组分和生长素的含量,检测限低至0.001.mg/L。本研究对样品的提取和纯化过程也做了改进,排除了杂质和色素对样品的干扰,为研究植物体内源激素对环境响应特征提供了有效的测定方法。

     

    Abstract: Plant endogenous cytokinins(CTK) and auxin(IAA) usually function at the same time,and thus a method that can detect these plant hormones simultaneously is of great importance in studying plant physiological processes.The contents of 6 endogenous CTK components and IAA in plant tissues could be determined simultaneously by HPLC.Re-sults showed that with reversed-phase column HibarRT250-4.6,at constant temperature of 45℃,and with methanol-1% acetic acid(40∶60,v/v) as mobile phase,the components could be precisely separated and determined at a flow rate of 0.6 mL/min by isocratic elution and with a UV detector at 269 nm.The detectable limitation of this method was as low as 0.001 mg/L.Steps of the extraction and purification of samples were also improved to avoid the disturbance of the impurity and pigment on the sample.These suggested steps provided an effective way to study plant hormones responding to environments.
