长期施用不同类型有机肥对赤红壤AMF多样性及功能的影响

Effects of long-term application of different types of organic fertilizers on arbuscular mycorrhizal fungal diversity and function in lateritic red soil

    目的 探究长期化肥配施有机肥对赤红壤甘薯土壤中丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)多样性和群落组成的影响,揭示其对甘薯产量及养分吸收的贡献。
    方法 依托2007年在福建省莆田市开始的花生−甘薯轮作长期施肥定位试验,在试验第16年(2023年)甘薯收获后,选取不施肥对照、单施化肥以及化肥配施商品有机肥、猪粪、秸秆5个处理,采集表层土壤样品,测定土壤化学性质和AMF群落组成,调查甘薯产量及养分吸收量。
    结果 与不施肥对照相比,长期单施化肥显著提高土壤有机碳及全量、速效养分含量(全钾除外),但土壤pH由6.29显著降至4.74,导致土壤酸化;同时,AMF群落物种丰富度和谱系多样性均显著降低,而球囊霉属相对丰度有所增加。与单施化肥相比,配施有机肥不仅缓解土壤酸化,还进一步提高土壤有机碳及氮、磷养分含量,其中猪粪处理效果最佳,土壤pH回升至6.12。此外,配施商品有机肥和猪粪能够维持AMF群落的物种丰富度,配施3种有机肥均显著提升了AMF谱系多样性。与单施化肥相比,配施3种有机肥均提高了3个AMF优势菌群中硬囊霉属的相对丰度,并降低了球囊霉属的相对丰度,猪粪处理还显著富集了球囊霉科未定属。网络分析表明,配施不同有机肥均显著改变了AMF共现网络结构,商品有机肥提高了网络复杂度,猪粪提高了网络连通性,而秸秆处理表现出更高模块度。LEfSe分析显示,球囊霉属VTX00400和球囊霉科未定属ASV78分别是商品有机肥和猪粪处理的特征类群。与单施化肥相比,配施商品有机肥、猪粪、秸秆处理甘薯产量分别提高11.39%、30.70%、24.05%。相关性分析表明,球囊霉科未定属与甘薯氮、磷吸收量显著正相关,球囊霉属VTX00400与甘薯产量及磷、钾吸收量显著正相关。随机森林分析表明,土壤有机碳是影响甘薯产量和养分吸收的关键因子,球囊霉科未定属和球囊霉属VTX00400分别在一定程度上解释了磷、钾吸收差异。
    结论 不同类型有机肥对赤红壤AMF群落及其共现网络具有差异化调控作用。配施猪粪缓解土壤酸化,特异性富集与氮、磷吸收相关的球囊霉科未定属,提高土壤养分有效性,并通过降低网络模块度、增强跨模块联系,显著提升群落整体连通性和协同作用,有利于形成互利型微生物互作网络。商品有机肥较好地维持了AMF群落丰富度和网络复杂度,并富集与磷、钾吸收相关的球囊霉属VTX00400,既增强AMF类群间联系,又保留适度竞争与较高分类多样性,表现出较强的网络稳定维持作用。配施秸秆表现出较高的网络模块度,促进AMF群落形成清晰的功能分区,减少网络冗余互作并降低对原有群落结构的扰动。综上,猪粪部分替代化肥是赤红壤甘薯体系中兼顾土壤改良、AMF群落优化与作物增产的优选施肥措施。

     

    Abstract:
    Objectives This study aimed to investigate the effects of long-term combined application of chemical and organic fertilizers on the diversity and community composition of arbuscular mycorrhizal fungi (AMF) in lateritic red soil under sweet potato cultivation, and to elucidate their contributions to sweet potato yield and nutrient uptake.
    Methods The study was conducted based on a long-term fertilization experiment established in 2007 in Putian, Fujian Province, China, under a peanut-sweet potato rotation system. In the 16th year of the experiment (2023) after sweet potato harvest, five treatments were selected: no fertilizer control (CK), chemical fertilizer alone (NPK), and chemical fertilizer combined with commercial organic fertilizer (NPK+CM), pig manure (NPK+PM), or rice straw (NPK+RS). Topsoil samples were collected to determine soil chemical properties and AMF community composition, and sweet potato yield and nutrient uptake were determined.
    Results Compared with the no-fertilizer control, long-term application of chemical fertilizer alone significantly increased soil organic carbon and total and available nutrient contents (except total potassium), but markedly decreased soil pH from 6.29 to 4.74, resulting in soil acidification. Meanwhile, AMF species richness (Chao1 index) and Faith’s phylogenetic diversity were significantly reduced, while the relative abundance of Glomus increased. Compared with chemical fertilizer alone, combined application of organic fertilizers not only alleviated soil acidification but also further increased soil organic carbon and nitrogen and phosphorus levels, with pig manure showing the strongest effect and increasing soil pH to 6.12. In addition, combined application of commercial organic fertilizer or pig manure maintained AMF species richness, while all three organic fertilizers significantly increased AMF Faith’s phylogenetic diversity. Compared with chemical fertilizer alone, all three organic fertilizer treatments increased the relative abundance of Sclerocystis among the three dominant AMF taxa and decreased that of Glomus, pig manure additionally significantly enriched unclassified_f_Glomeraceae. Network analysis showed that all organic fertilizer treatments significantly altered AMF co-occurrence network structure; commercial organic fertilizer increased network complexity, pig manure increased network connectivity, while straw treatment resulted in higher modularity. LEfSe analysis identified Glomus VTX00400 and unclassified_f_Glomeraceae ASV78 as characteristic taxa of the commercial organic fertilizer and pig manure treatments, respectively. Compared with chemical fertilizer alone, combined application of commercial organic fertilizer, pig manure, and rice straw increased sweet potato yield by 11.39%, 30.70%, and 24.05%, respectively. Correlation analysis revealed that unclassified_f_Glomeraceae was significantly positively correlated with nitrogen and phosphorus uptake, while Glomus VTX00400 was significantly positively correlated with yield and phosphorus and potassium uptake. Random Forest analysis indicated that soil organic carbon was a key factor influencing sweet potato yield and nutrient uptake, and unclassified_f_Glomeraceae and Glomus VTX00400 partially explained the variation in phosphorus and potassium uptake, respectively.
    Conclusions Different types of organic fertilizers exerted distinct effects on AMF communities and their co-occurrence networks in lateritic red soil. Combined application of pig manure alleviated soil acidification, specifically enriched an unidentified genus within Glomeraceae associated with nitrogen and phosphorus uptake, and enhanced nutrient availability. Moreover, by reducing network modularity and strengthening cross-module connectivity, pig manure significantly improved overall community connectivity and synergistic interactions, thereby facilitating the construction of a mutualistic microbial network. Commercial organic fertilizer effectively maintained AMF richness and network complexity, and enriched Glomus VTX00400 linked to phosphorus and potassium acquisition. This not only intensified interspecific linkages among AMF but also preserved moderate competition and high taxonomic diversity, reflecting a strong capacity for sustaining network stability. Straw incorporation exhibited the highest modularity, promoting clear functional compartmentalization within the AMF community, minimizing redundant interactions, and mitigating disturbances to the pre-existing community structure. In summary, partial substitution of chemical fertilizer with pig manure represents a preferred strategy for simultaneously improving soil properties, optimizing AMF communities, and increasing crop yields in sweet potato systems on lateritic red soil.
